No, although if you have rust in the cooling system, and bits of rusted out metal are flaking off and getting into the coolant, it can have other detrimental effects.

Put the cap back on the cooling system and take the vehicle for a short drive, or run the engine till the entire system is up to temperature and then check the oil for contamination.
