I have driven from Nogales, Arizona to Guadalajara many times, and my family travels back and forth a lot. In the old days before the toll roads were constructed, it was always a three day trip. The first night we always spent in Ciudad Obregon. The second night was spent in Mazatlan, and on the third day we arrived in Guadalajara. We never traveled at night. Never.
The advent of the toll roads saw the time from Nogales to Guadalajara drop a whole day. The first day went spent the night in Culiacan, which no one recommends. The second day we rolled all of the way into Guadalajara in the mid afternoon.
No one stays in Culiacan, except we did. Do not spend the night in Culiacan like we did. We could hear gun fire outside throughout the night while in our high rise hotel. Again, not recommended. Keep away from windows and low to the ground in your room. (Culiacan is not as bad as this answer says, you make it sound like you´re in the wild west. Although this city is known for being the cradle of drugdealers in Mexico, it is a friendly city and as long as you follow common courtesy rules you should not have any problems.
A better choice would be stopping in Navojoa the first night, and then making it into Guadalajara in the late afternoon the second day.
The toll road all of the way to Guadalajara from Nogales cost about $450.00 US. (Actually it´s only $150.00 US. I can prove it I still have the receipts) Not cheap anymore. I think I will take the old road soon to see how it is getting along. Out of about six times up and down that route, only one was on the new toll road. I may be living dangerously, but I am cheap, too.
The toll road was two lanes all of the way down and in good repair. Hence, the shortened time to Guadalajara. If you are in a hurry, take the toll road. I should probably advise everyone to take the toll road, no matter what.
My sisters are trying to talk me out of driving the old road in October as I write.
