You can tell the difference based on their genitals. A male would have the penis and testicles combo while the female will have a vagina. If the male has been neutered the testicle sack will still be there.

If it is a short distance apart, you have a female.Also, a male will have a "hole", females will have a "slit" Always double check with a vet, as spayed/neutered cats or cats under 6 weeks old can be very difficult to identify.
