pop it off with a screwdriver then get a large socket about the same size and lightly tap it till its in

I suppose it is very possible to cause an unintended problem with the transmission while replacing the rear seal as it involves either removing the transmission, or at least pulling it back. If its an automatic transmission, you would have to remove the torque converter and flex-plate. If something doesnt get properly torqued to specs, then Yes you could cause a huge problem. You also run the risk of damaging the drive-line, slip yoke, Universal joints, etc.... Not the best job for a first timer to do in my opinion.
The rear section of the driveshaft has a slip yoke in it. This allows the rear section of the driveshaft to change length as the rear axle moves up and down. If this slip yoke binds, the change in length is absorbed by the slip yoke at the back of the transmission. Because the carrier bearing itself is fixed to the driveshaft, and the hanger that holds the bearing is fixed to the frame or cross member, when the rear slip yoke binds, it moves the entire driveshaft forward taking the carrier bearing with it, and pushing it out of the rubber in the hanger.
the purpose of the slip yoke is to allow for movement over bumps and different terrain. if there was no give ,you would constantly be trashing the driveshaft or transmission

A slip yoke is when the end of the drive shaft in a RWD vehicle plunges in and out of [most likeley] transmission when rear suspension is flexed.
I'd change the fluid and filter, but no guarantees.
It depends on how much fluid leaked from your transmission. 2 quarts would probably be considered ok. anything more, the transmission might start to slip intermitantly and cause internal problems. Mac's Transmissions Houston, TX 281-743-1000
Your transmission might slip on your ML320 because the transmission fluid is low or because the transmission is damaged. The most common cause of transmission slip is wearing of the transmission.
Lining worn to to improper use? Rear engine seal leaking on clutch? Improper adjustment?
