If you're working on a Mac, have you enabled Parental Controls under System Preferences recently? I had the -50 issue, and this was the cause.

temp workaround ... type in a search term and iTunes may access the store, then will act as normal. Until this bug is fixed, you may have to do a search every time on startup. jag
It is a software bug, most people will have to wait for apple to fix it
