Ears and eyes are often a good thing to look at. If the eyes are particularly runny/watery, if there's any discharge, or if the third eyelids of the kitten are showing, there is a chance that the kitten is feeling ill. Kittens often have bags under their eyes as their eyes begin to open.
Make sure that the ears do not contain ear mites. This will look like dark brown gunk in the ears, and will cause serious irritation. When a cat has ear mites, it will continue to scratch like a child with chicken pox- even if it scratches its own ears bloody.
Keep an eye out to make sure that your kitten is not vomiting, and that it does not have diarrhea. Also contact your local vet about medicating your kitten for worms, as kittens very often have these.
Be sure to have your kittens vaccinated. A combination/first vaccine is often given between 6 and 7 weeks, and a 'booster' vaccination given one month later. Contact your vet about additional vaccinations that you may want to get for them.
When a cat has a illness, most of the time it is fairly easy to recognize. The behaviour may change, it may stop eating/drinking, not active, vomiting, loootsss of hair falling out, bare patches in skin, yellow/ dry tongue, limpy tail, strange eye behaviour, etc.

You can't tell if they will be fluffy or not. As the kitten gets older it will get easier to tell if the kitten is longhaired or shorthaired, but in very young kittens it is very difficult to tell.
