I have a 92 4Runner and once you open the tailgate, there should be a piece of cloth covered cardboard that is on the inside of the tailgate. you can get a flat head screwdriver or use your hands and just pull it off. Only thing hold that panel in place is some plastic clips, they might break cause of the age of the vehicle, but are very easy to find.

The interior panel pops off and there is one screw behind the little panel underneath the strap handle.
Removing the inside tailgate panel from a 2001 Toyota four runner is a very easy process. The first thing that needs to be done is to remove the door handle. When this is done, you will see two christmas tree fasteners that need to be undone. This will allow you to just pop the panel out now. Once this is done, simply move the panel past the door handle and the panel behind can be easily removed one all of the fasteners are undone.

Remove the rear interior panel. It will simply pull away with a little force. You will need to remove the Philips screw on the rear seat catch and the bolt on the rear seat belt first. It is also a good idea to remove the molding under the tailgate. The speaker will be located behind the built in grill. FYI, Toyota uses a power speaker with built in amplifier. Best to replace with same.

By removing some screws and/or bolts.
On my 89 4Runner it is under the kick panel on the driver's side.
