If its from the airport Bear in mind that it may take you an hour to get across Mexico City from the airport (which is truly a horrendous journey). and then a minimum 5 hours to Guadalajara. I would recommend staying in Mexico City the night and recharge your batteries to do the whole trip the following day. The only sizable place to stop en route would be Toluca and you have dismissed that.
