The amount of sand depends upon the design of your waterscape. I always have at least 3 inches of sand at the back and slope the substrate towards at least 1 inch deep at the front of the tank.
You do not need gravel in a tank if you choose not to use it. The amount depends on the look you want to achieve.

I'm sorry but no turtle can live in a 2.5 gallon tank. You will need a least a 20 gallon tank and that is even iffy when they grow up I would say get a 30-40 gallon tank for one turtle.
