It depends entirely on the project - Photoshop is an image and graphics creation and editing tool, based on pixel files. Illustrator is an original artwork and design program that is based on vector files. The difference is that vector files can be resized without any loss or change in resolution. The easiest way to view the 2 is to imagine Photoshop as a digital tool for photographers and artists creating digital images, and Illustrator as a digital program for painters, sketchers, print artists, etc.
Both programs are powerful in their own respect, but only in respect to the project at hand. For example, Illustrator is great for making print brochures, magazine layouts, original art, etc., but it can't edit photos or similar images. Conversely, Photoshop is the program if you're looking to work with digital images, but it's not for making print publications.
I would recommend Adobe Photoshop or Adobe Illustrator.
Vector graphics. Photoshop can work with vector graphics, but Illustrator is ideal.
Adobe illustrator (AI) is a vector file format to save vector illustrations. It is the default format for Adobe illustrator. Photoshop cannot save the images in to AI format but it can be opened in Photoshop.

Use Adobe Illustrator to create your graphics, then use Adobe InDesign for the layout of the flier and the typographic design. You can place your graphics from Adobe Illustrator directly into Adobe InDesign. If you are using photographs instead of creating illustrations, use Adobe Photoshop to edit your photos. However, do not attempt to do the layout and typography in Adobe Photoshop - use Adobe InDesign for that.You could also do everything, including the layout and typography, in Adobe Illustrator. If you decide to use a mix of photos from Adobe Photoshop and illustrations from Adobe Illustrator, use Adobe InDesign to do the layout. You have options with Adobe. Amlove has a good answer. But a single-page flyer? Like a handbill or something? I think I'd just work up the photos in Photoshop, then do the layout directly in Illustrator. Flyers are pretty much pure display type, and you can really doctor up type in Illustrator.
