No, I'm afraid that's another one of those old wives tales I'm afraid.
Most pups will yelp when you pick them up by their scuff, it is their natural way of communication with their parents on how much pressure is applied when picking them up, before hurting them. Alot of pups do it to protest aswell, especially when they have their mind set on doing something else.
