Demographers use a "dissimilarity index" to determine the relative segregation of various metropolitan areas. A score of 0 means there is complete integration and a score of 100 means complete segregation (distinct ethnic neighborhoods). The level of segregation in various studies only looks at white/black segregation.
These have concluded that metro Milwaukee is the most segregated metropolitan area in the US with a score of 81.5. (Note that this is the metropolitan area, not the city. Studies on segregation don't seem to rank individual cities).

The Tuskegee airmen were the first African-American Military Aviators to serve in WWII. This was before the time of Dr. King, and the Jim Crow Laws where still in effect. During WWII, the Military, like many branches of the US Government, was racially Segregated. They where subject to discrimination inside and outside of the US Military. They where known for training and flying with Distinction, despite the discriminating times of the 1940s.

As this question clearly calls for an opinion, my answer is Brown v. Board of Education which held that racially segregated public schools were in violation of the equal protection clause of the US Constitution, thus directly overturning Plessy v. Ferguson. ----
