Most of the time they don't "mean" anything.
When I first got my license in Missouri, by comparing it with that of my friends I was able to figure out that the first letter of the license number was the first letter of my last name (this was pretty obvious), and it also seemed to be true that the next two digits were the numerical value of the second letter of my last name (in my case this was U, so the digits were 21).
Missouri no longer issues licenses using that numbering scheme any more, and like most other states that I'm aware of they just use your social security number if you have one and don't specifically object to its use, or a semi-random number they generate if you do object or don't have an SSN for some reason.
The numbers running down the side of the drivers license is an internal code from the DOL. It does not mean anything specifically it is just a sort of identification code the DOL uses.
