The eastern part of the Roman Empire had a mix of cultures.

The culture of the portion of the eastern part of the Roman Empire around the eastern Mediterranean was Hellenistic; that is, it was influenced by the Geeks.

Between 334 BC and 326 BC Alexander the great conquered the Persian Empire, which stretched from Turkey to the river Indus in Pakistan and extended into central Asia. It also covered Iraq, Syria, Lebanon, Israel/Palestine, and Egypt. After Alexander's death there were fights between his generals and eventually his conquests were partitioned into the kingdom of Pergamon (in western Turkey) the Seleucid Empire (which was centred on Syria and covered most of the rest of the Asian territories) and the Ptolemaic kingdom (Egypt, eastern Libya, the eastern part of the Mediterranean coast of Turkey and Cyprus). Therefore, these areas were ruled by Greek dynasties and were heavily influenced by Greek culture. The cultures of the local peoples remained. Greek culture in these areas was influenced by the local cultures, particularly in Egypt. The biggest upholders of their culture were the Jews, who clashed with the Greeks in Judea.

When the Romans conquered Greece (in 176 BC) and later the rest of the eastern Mediterranean they were already Hellenised. They were deeply influenced by the Greeks. Roman rule brought a degree of influence by Roman culture and the influence of Roman imperial ideology.

In the Balkan Peninsula (south-Western Europe) there were the cultures of the local ethnic groups: the Illyrians (in the former Yugoslavia) the Dacians (in Romania and Moldova) and the Thracians (in Bulgaria and European Turkey). These groups were heavily romanised. So much so that the language of the Romanians (the descendants of the Dacians who were not later conquered by the Slavs) is a romance language; that is, a language descended from Vulgar Latin (a mixture of Latin and local languages).

In what way did the emperors of eastern roman empire differ from the emperors of the western roman empire?

There was no such thing as an eastern Roman Empire and a western Roman empire. It was a Roman empire. Period. Historians are the only ones who denote eastern and western, but they do this for clarity when speaking about certain sections or aspects of ancient Rome. Now, as for comparison between the eastern and western sections of the empire, the eastcrn section was wealthier than the western and consequently lasted longer than the western section and the eastern section was more heavily influenced by the Greek culture than the western part.
