There isn't a really adequate answer to that. None of the unusual words - like "supercalifragilisticexpialidocious" or "Floccinaucinihilipilification" count, as they are used often enough as examples.

Literally the least used would be one that was used not at all, and there are thousands of examples of those.

The field in which they would come from would be some obscure branch of taxonomic science, where it was the latinate name for some rare bug or plant that no one cares about. Or some chemical compound with no application.

If one wishes more pure English, and not simply some rare scientific term, one would look amongst the obsolete words that we no longer use, such as "abatude", in which one cuts a portion of a coin out, thus devaluing it.

If one is wanting the least used word that is not scientific or obsolete, and further, still actually used, then you would still be out of luck, because there would be thousands of ties for least used. And really, no meaningful way of determining which is "the" least used.

One study ranked the most commonly used words, and at number 86,800 was "conquistador". However, there are several hundred thousand other words that weren't listed.

Myself, I would offer - with no proof - the word "cwm". Pronounced "koom", it means "from the valley". The only time I'm aware of it being used is in a holoalphabetic sentence that most people don't bother with. "Quartz glyph job vex'd cwm finks." contains every letter of the alphabet once and only once.

So that word is used, but is certainly amongst the "least used". Perhaps it might be fairest to say that I offer that as "the word that is least used amongst words that are still used."
