That lock is to keep someone from stealing your spare tire. When you remove the lock then you will be able to slide the tool in the hole and let the spare tire down to the ground.
First, remove the two bolts at the top of the light cover and the tab that holds the bumper closest to the headlight. Then, remove the headlight. The bulb will be held by a twist lock. Finally, place the new bulb into the lock, replace the headlight, put the bumper tab back in place, and screw the two bolts back into place.
