In Shakespeare's plays, the major difference between a tragedy and a comedy is that the main character dies in a tragedy. A Shakespearean comedy can be very tragic and depressing, but so long as the main character remains alive, it is not actually a tragedy.
The main character in "An American Tragedy" is Clyde Griffiths, a young man who aspires to a better life but becomes entangled in a love triangle that leads to a tragic outcome.

But actually, we view any story in which something sad happens to a character we care about as being tragic. If the character who we care about happens to be the main character of the story then the story is a tragedy.
