This is a very interesting question, as it could have two different answers.

On one hand, we have DNA, which is important for obvious reasons.

On the other, we have ATP, which provides energy for every cell on the planet.

Since there are other ways to store genetic information besides DNA, I would deem ATP as the most important molecule in the human body, as nothing can survive without energy.

To the extent of a layman's understanding, the macro-molecule that has absolutely the most import to life may very well be that of water. Following behind and "running" a close second may very well be macro-molecules of proteins; all genes in the genomes of organic life contain the "blueprints" for synthesis of proteins.
