Take the Uptown 2 or 3 (of the 1-2-3, the red line) from Fulton Street to 42nd Street-Times Square (at 7th Avenue). The 2 and 3 are express trains, and it's only 4 stops from Fulton Street to Times Square: Fulton Street ---> Chambers Street ---> 14th Street ---> 34th Street-Penn Station ---> 42nd Street-Times Square.

There are 4 different Fulton Street Stations, although they are all connected by underground tunnels so you can transfer between them without exiting the subway system. There's a Fulton Street Station on the 2 and 3, the A and C (of the A-C-E, the blue line), the 4 and 5 (of the 4-5-6, the dark green line) and the J-Z (the brown line).

The Fulton Street Station on the 2 and 3 is at the intersection of Fulton and William Streets. To get there, walk 6 blocks west on Fulton Street to William Street.

You should be able to find Fulton Street easily. It's at the north end of the South Street Seaport (which is only about a block long, from John Street to Fulton Street). Fulton Street is a narrow, cobblestoned street (at least, the first block of it is), and the South Street Seaport Museum is located on that first block. You'll see the Heartland Brewery on the corner, and an Ann Taylor store next to it.

The 5 of the 4-5-6 stops at Grand Central Station. If you take the 5 to Bowling Green, it will drop you off pretty close. From there just keep walking southeast (the streets down there are crooked, so it'll seem like you're walking straight south) until you hit the water.

You can also take the J or Z of the J-M-Z (the brown line) to Broad Street, or the N or R of the N-R-Q-W (the yellow line) to Whitehall Street/South Ferry. Same walking directions.

Without a more specific starting point, this question is unanswerable. "Upper Manhattan" could refer to anywhere in Manhattan above 59th Street.
