Although the problem could be caused by a bad spark plug, that is not the whole story. First of all which engine are we talking about here? The 5.4L V-8 "Triton" engine has coil-on-plug (COP) assemblies, which eliminates spark plug wires, so the idea of replacing spark plug wires on a 5.4L engine is not an issue. If you have a 5.4L V-8 engine, then more likely problem is a bad coil, and it's fairly easy to replace the #5 COP assembly, and related spark plug, since it's located at the front of the engine, on the driver's side. A new COP and plug will cost about $50 at your local auto parts store - try that route, and likely the problem will be resolved.
An engine cylinder misfire has been detected in cylinder # 1
A misfire is when one of the cylinders is accelerating without actually doing anything, it's just a dead-cylinder. In order to fix this you need to tighten the cylinder or adjust the engine connector.

Could be a bad ignition coil each cylinder has its own
Most likely you have a misfire caused by an intake manifold gasket leak at #1 cylinder. That or basic misfire stuff like spark plugs or ignition wires. Check the intake
It means that more than one cylinder has a misfire history. PO301 would be cylinder number 1 misfire. PO302 would be number 3 cylinder misfire.
An engine cylinder misfire has been detected on the # 1 cylinder and the # 8 cylinder firewall 4 - 8 3 - 7 2 - 6 1 - 5 front of Ford Expedition
I had the same problem with a 1999 Expedition and it was a bad plug. Ford had a problem also with the plugs blowing out. It cost me about 1200.00
A backfire is caused by a misfire... a cylinder fails to ignite the fuel which may cause a stutter, it may also cause a back fire due to it pushing that un-ignited fuel into your manifold where it then ignites from the heat of the other cylinders and boom you have a backfire...
