If you are referring to Mark Antony, he never actually ruled Rome. He was part of a three-man
alliance which was called triumvirate, with Octavian (later he was called Augustus) and Marcus Aemilius
Lepidus which fought against the assassins of Julius Caesar. After they defeated them, Octavian took charge of the western part of the empire (including the city of Rome), Mark Antony too charge of the east and Lepidus was in charge of northwestern Africa and Spain. Octavian then declared war on Mark Antony and his ally, Cleopatra VII of Egypt. Octavian defeated them and they committed suicide. Mark Antony was in change of the east from 40 BC to 30 BC.
Mark Antony, it should be noted, never actually ruled Rome for long. While Caesar was away in the east, Antony was his Master of the Horse and ruled and was responsible for the city of Rome. He was also the supreme authority after Caesar's murder, as he was the remaining consul and brokered the "deal" with the conspirators.
After Caesar defeated Pompeii and was dictator, Marc Antony was Caesar's Master of the Horse, which was the one who ruled Rome in the dictator's absence. Caesar appointed him counsul in 44 BC. and after Caesar's death he was the man in charge.

No his ex-wife did, but it only led to disaster. Marc Antony originally ruled Rome as Caesar's Master of the Horse while Caesar was in the East going after Pompey. He had a problem with unrest and came down hard on the rioters, incurring the anger of Caesar. When things were patched up between the two of them, and Antony was counsul with Caesar, Caesar was killed and Antony was the sole authority in Rome.
Antony just wants the support of Brutus' followers so he can have complete control over Rome.

Cleopatra was not so much important to Rome as she was to Octavian. She was the tool that Octavian was able to use against Antony and to wage the war against him. By accusing Cleopatra of corrupting Antony, Octavian was able to get the senate to declare war against her. In reality this war was against Antony for sole rule, but Octavian had to avoid all semblance of a civil war and Cleopatra and her conduct with Antony was the perfect opportunity for him.
