It first started with a short made while the current creators, Trey Parker and Matt Stone, were in college. The short was stop-motion animated and entitled "Jesus vs. Frosty". It featured very similar characters and settings to what is seen in the show. Years later, another similar short entitled "Jesus vs. Santa" was made, which launched internet fame and eventually got Comedy Central to pick the creators up to make a pilot episode for South Park.
