In Manhattan, the only direct transfer from the E to the R, or the R to the E, is at 42nd Street. Take the E to 42nd Street-Port Authority Bus Terminal, and then walk through the tunnel that connects the 42nd Street-Port Authority station to the 42nd Street-Times Square station, and get the R at Times Square.
There are a number of ways to indirectly transfer from the E to the R. You can take the E to West 4th Street, transfer there to the Uptown B-D-F-M to 34th Street-Herald Square, and then transfer to the R at Herald Square.
