A Papal bull and the Treaty ofTordesillas between Spain and Portugal of 1494 was put into effect which established theSphere of Influence of each nation within the new world. While the geography of the new continents had not been adequately mapped out at the time, this division generally became effective.

As a result of the Treaty of Tordesillas the Spanish received the land west of the line and Portugal east of the line. That is why most of South America speaks Spanish and not Portuguese. Since the Pope drew a line down the center of the Atlantic Ocean, and what is present day Brazil sticks out, it essentially became the only thing the Portuguese controlled in the New World.

Does South America's population remains concentrated in its interior?

I believe most of the population of South America is located at or near the coastal regions (seacoast primarily) in larger cities and towns, some populations live along large rivers and in high mountains, but these are not the primary population concentrations of South America. I believe most tribal populations will contribute to the strong tendency of statistical migration to the coastal cities, and will not grow to large sizes in the interior regions, in the present era and foreseeable future.
