I have a 1998 Isuzu Amigo and ran into this problem after installing a new intake gasket. I purchased a Hayes repair manual and used their diagram to reconnect the plug wires and found out that it was incorrect. I have also seen the same diagram on the internet. The books states the firing order on the coil is 1-3-4-2 from left to right but is actually 1-4-3-2 from left to right facing the engine. Hope this helps.

Could be a bum fuel injector. Pull number 6 spark plug again, if it is wet you have a spark problem. If the plug is bone dry you have a fuel problem, probably a bad injector. Swap injector with another cylinder and see if the misfire switches cylinders or remains on number 6 cylinder.

Think a little deeper. Combustion that fails is a misfire. You have addressed the spark side of it. There is a fuel and compression possible trouble. Try swapping the fuel injector for that cylinder with another and see if the misfire moves to the other cylinder. Compression test the cylinder to see if it is in specs. Try a cylinder leak down test and see if it is losing compression. Perhaps there is a carbon build up on the valves preventing good seating?
