It could either be the battery inside the remote.... which I'm sure you've tried. Or your sensor inside your car could have gone bad. Depending on what kind of car you have, it could turn out to be quite epensive to fix. I'd suggest the old fashioned way of using the actual key.

If you rule out the more serious problems, of something wrong with the alarm, or the remote itself is defective, then it sounds like the remote needs to be programmed.On a 2001 Volkswagen, this is a process that requires two keys.Insert master key into the ignition lock & switch on the ignition (do not start car).Using second key lock the vehicle from the outside manually (use key in door lock).Press unlock button on the remote control one time, wait at least two seconds or more.Manually open car using key in door lock.Remove key from ignition.Test your remote.

Assuming that your other doors lock and open properly check the solinoid for the drivers door.
Sounds like you need to replace the drivers door lock actuator.

If the remote locks the door properly that tells me that the wiring and the lock solenoids are operating OK. I'm also taking it for granted that the switch you spoke of is the door lock switch on the drivers door. Does the passenger door switch work properly? If so, then you probably have a bad switch on the drivers door. (IMPROVE) You can remove the door lock mechanism from the panel and gently pry open. Carbon build-up is common on the switches. Use a pencil eraser to remove the carbon and ensure the switch functions properly. Re-assemble.
No unfortunately you cannot. an 05 remote will be in no way compatible with a 95 door lock reciever.
