Yes but, if you relieve the pressure by opening the radiator cap (when engine is cold) then replace the radiator cap before removing the knock sensor, only a small amount of coolant will come out while you change the unit.

You have another problem other then the knock sensor if the engine will not start and run. A knock sensor will not cause the engine not to run.
The knock sensor is basically a one inch thick washer with a sensor plug on it. it will be attached to the block with a bolt that just runs through the center of it The knock sensor module is located inside the little cover on the VCM.
Overheating, yes. Loss of antifreeze, no.
