Because negative staining requires the use of an acidic stain, which will not penetrate the cells because of the negative charge on the surface of the bacteria. As a result, the unstained cells can be easily identified against the colored background.
Methylene blue can be used to prepare slide for animal cells. For example, if you want to examine a piece of your cheek cell, you would take a piece of cotton wool and rub it on the inside of your cheek and the rub it on a microscope slide, the you add a little distilled water and then a drop of methylene blue. The final step you would take, is to cover the slide with a cover slip, then place it under the microscope.
When you added methylene blue to the living yeast cell, the methylene blue is actually a dye so you could see the cells, therefore, the yeast cell became blue. It is much the same with hair dye. If you have a bottle of blue hair dye, it will dye your hair blue.
Methylene blue works in several ways.?æ It works as a bladder anti-spasmodic, as a stain to make cells more visible, as an oxidizing agent, can be used to treat psoriasis, and as a placebo by doctors.?æ?æ
If it has a use by date you are supposed to observe that date. I personally would ignore the use by date and use it
It removes the crystal violet (primary stain) from gram negative bacteria. It does not remove Crystal violet as easily from gram positive bacteria, because the highly peptidoglycan walls of gram positive bacteria interact with crystal violet and iodine to form a strong bond (CV-Iodine complex). So, gram positive cells hold on to the stain instead of letting it wash away.

Immobilizing the bacteria on a slide prior to staining helps prevent them from being washed away during the staining process. This ensures that the bacteria remain in place and are properly situated for observation under the microscope. It also helps to maintain the integrity of the sample and improve the accuracy of the staining results.

When handling methylene chloride, it is important to wear protective gloves, goggles, and a respirator to prevent skin contact, eye irritation, and inhalation of harmful vapors. Work in a well-ventilated area and avoid using the chemical near open flames or heat sources. Additionally, store methylene chloride in a cool, dry place away from incompatible materials.

Try not to get the negative credits in the first place. In other words, behave!
You remove the negative sign long enough to divide the numerator by the denominator then put the negative back on its place value
We suggest painting the surface of cured concrete with either an appropriate stain or paint available at your local "do it yourself" type hardware store (such as home depot or lowe's). You can even purchase a rolled stencil to place over the surface of the concrete and will result in a finished appearance of flagstone or pavers!
