Most people think it was Pilgrims and Puritans, but there is evidence some small settlements were established before that. One historian (Robert J. Allison) suggests that there were ships that arrived in Massachusetts as early as 1617. In 1624, a gentleman named Samuel Maverick built a trading post near what is today called Boston Harbor; in 1626, William Blackstone arrived and tried to set up a settlment in Wessagusset (today Weymouth); he remained in the New World after the settlement did not survive, and he was still living not far from Maverick's outpost when the major European excursions to Massachusetts began in 1630. But I guess the short answer is that the first permanent settlement was founded by John Winthrop-- he was a British lawyer who led his expedition to establish a religious settlement, a "community of saints," a "city on a hill" that would be a model for other cities in the New World. He became the first governor of the Massachusetts Bay Colony, and is thus credited with having set up the first permanent settlement, although the other two gentlemen seem to have had little outposts...
