There are special drills that are made specially for drilling many types of glass.
They work perfectly well on mirrors. For the best results you should drill from the back of the mirror, this reduces the chance of the reflective coating from splitting away from the glass.
Expensive door jams have screw holes for tapcoms to install into concrete foundation or wood framed jams. Cheap door, you have to morter the jam in place. I drill a 1/2in hole through the rise plate(frame stop ) then drill hole size of screw through the outer plate that is against the foundation or jam. This way you do not see the screws that are holding the frame.

I think the correct way to do this is to mark out where the hole is required, get some putty and form a cupped mound below, fill with varsol or white spirit, and drill the hole slowly with a special glass drill bit. You must keep the bit lubricated with the liquid to dissipate heat and prevent the mirror from cracking.
