I hyave a 2000 Toyota Sienna CE. When the first time this happened to me I took it to the dealer and got a charged a whopping $300 (includes part). So when the part broke again, I decided to do it myself. From the inside of the vehicle, carefully remove the plastic panels. Do this carefully as not to break the plastic fasteners. After this, locate the broken part. From the inside you can open the door now by operating the door mechanism. Once you open the tailgate, remove the part and replace. It's better to remove the part first then take it to your dealer as not to make a mistake and get the wrong part. After replacing the part, just work backwards to install everything. Note: Be sure to take photos of the part before taking it off, so if you forget you can refer to the photos.

First you need to remove the trim bezel around the handle on the outside of the tailgate. Then remove the three bolts on the inside of the tailgate,that hold the handle in place. Next remove the latching mechanism rods from the handle,and remove the handle from the tailgate.
should be an access panel on the inside of the tailgate for repairing the hardware

The inside of the tailgate, that is the part that faces up when the tailgate is down will have a panel that is held on by screws, it will give you access to the inner workings. Usually it is a little 20 cent retainer that has broken and is available at any parts store.
Happened to me a few months ago. Remove the inside rear panel and pull the latch manually. Then you can unscrew the broken latch and replace it.
Don't know if this will help, but I had the same trouble with my 2002 and my mechanic had to take apart inside panel on back tailgate and there was a switch in their that had broken. The part was just as expensive as the labor and it was quite a task.
If the tailgate is dropped hard it can toggle the locking mechanism. If you don't have a key to unlock the tailgate, you'll have to remove the plastic side of the tailgate (the inside) and flip the locking lever back to the unlocked position. While you have the cover off you might as well put some drops of oil on all of the metal moving parts inside the tailgate.
Seperate valve for outside faucet turned off, broken valve stem inside of outside faucet or handle stripped at faucet.
