Not necessarily. A state's capital city is not always the most populated area of the state. In fact, the most populated city in a state is usually NOT the state capitol. Many (but not all) capital cities were the most populated in their state at the time they joined the union, which was over 100 years ago for most states. Over time things change.

NYC is its largest city in New York, while the capital is Albany.

Alaska capital is Juneau, not Anchorage or Nome

Texas capital is Austin, not Houston or Dallas

California capital is Sacramento, not Los Angeles or San Francisco

Pennsylvania capital is Harrisburg, not Philadelphia

Illinois capital is Springfield, not Chicago

Michigan capital is Lansing, not Detroit

Florida capital is Tallahassee, not Miami

Minnesota capital is St. Paul, not Minneapolis

Montana capital is Helena, not Billings
