Mary I was the daughter of King Henry VIII and Katherine of Aragon, and, therefore, heir to the throne of England. Mary Queen of Scots was the daughter of King James V of Scotland and Mary of Guise. She was Queen of Scotland and, very briefly, France. She was actually Mary I of Scotland but the name Mary Queen of Scots is generally used to differentiate her from Mary I of England.

Yes, they are different women. Mary I (1516 - 1558) nicknamed "Bloody Mary" for her persecution of Protestants during her reign, was the daughter of Henry VIII (1491 - 1547) and his first wife, Catherine of Aragon (1485 - 1536).

Mary, Queen of Scots (1542 - 1587) was the grand-daughter of Margaret Tudor (1489 - 1541), Henry VIII's older sister. Margaret Tudor had married James IV of Scotland, and so Mary Queen of Scots came to inherit that throne.

No, Mary I of England was King Henry VII's sister. Mary, Queen of Scots was not associated with England until captured by Queen Elizabeth.
