if this happens then you can easily wash away your bacteria. the heat fix kind of makes the bacteria stick to the slide

What is the influence of the pH on the gram reaction in gram staining?

In what regard? You need heat in order to heat fix the bacterial cells to the slide. This adheres cells to the slide. Otherwise, the bacterial cells would wash off the slide during the Gram staining process. If you leave the slide in the Bunsen burner too long, then you can distort the bacterial cell shape and size and also have other artifacts appear on the slide that are not bacterial cells.

How would you define a properly prepared bacterial smear?

the bacteria are evenly spread out on the prepared slide in such a concentration that they are adequately separated from one another bacteria are not washed off the slide during staining bacterial form is not distorted

During the performance of the simple staining procedure you failed to heat fix your EColi smear preparation Upon microscopic examination how would you expect this slide to differ from correct slide?

The bacterial smear will wash away during the staining procedure. This is avoided by heat fixation, during which the bacterial proteins are coagulated and fixed to the glass surface.

Purpose of fixing the bacterial film?

The purpose of heat fixing the bacteria to the slide is so during the gram staining procedure the bacteria doesn't wash off. If you didn't heat fix the bacteria to the slide, it would not stay on the slide.

What is thee purpose of adding methylene blue to the specimen?

Adding methylene blue to a slide will stain animal cells and make the nuclei more visible.

Give the reason for passing bacterial smear through the flame before staining?

It dries the smear and fixes the cells to the slide

What would happen if the step involving Gram's iodine was inadvertently left out of the Gram staining procedure?

Gram staining refers to separating bacterial species into two groups, gram-positive and gram-negative, which defines the physical and chemical properties. Skipping a step in this process could result in a mixed group of the bacterial species.

What is the importance of heat fixing the slide before staining?

You heat fix a slide by passing it through a blue flame a couple of times (with th cells facing up). you do this to denature any enzymes that might lyse the cells or interfere with the staining procedure. you also use it kill the organism and to adhere the organism to the slide for staining
