peptone iron agar is used to demonstrate the production of hydrogen sulfide. ıt is more sensitive than lead acetate agar. some organisms have the enzyme to liberate hydrogen sulfide from sulfur containing organic compounds-such as protein-. when H2S is liberated by the enzyme,, the sulfur of the H2S combines with the iron in the PIA medium to form an iron sulfide. The iron sulfide is manifested as a black precipitate in the medium. so you can use in the separation and identification of bacteria. Peptone glucose agar (PGA) medium
Beef extract for carbohydrates Peptone for protein and NaCl as salt
Modified nutrient glucose agar is composed of agar, water, glucose, extracts, peptone and sodium chloride. Modified nutrient glucose agar allows organisms to grow.
MacConkey agar contains bile salts, lactose, peptone. crystal violet dye, and neutral red dye. The agar appears a medium pink with a copper undertone.
Because the peptone iron agar is used to detect ANAEROBIC bacteria. If you stab it deep into the agar you allow the bacteria to grow in the absence of oxygen. If you only inoculated the surface the bacteria wouldn't grow.

peptone, yeast extract,meat extract..
Peptone and beef extract
Peptone is the simplified source of protein and can be readily uptake by the microorganism. In nurtient agar it is the basic component/nutrient after which beef or yeast extract is used which are supplementary nutrients.
Peptone glucose agar (PGA) medium
thats a good question but unforunately i dont now the answer
Beef extract for carbohydrates Peptone for protein and NaCl as salt
sabourauds agar contains the following : peptone : 1 g glucose : 4 g agar :3 g d/w : 100ml PH : 5.4
