As of 2013, there are four: Dover (DE), Jefferson City (MO), Juneau (AK), and Pierre (SD). Most of them are at least served by a major highway that connects to the interstate system, but Juneau is surrounded on land by mountainous terrain and can only be reached by sea or air.

Many people think Honolulu does not, but in fact Hawaii does have highways that are part of the interstate highway system, though they obviously do not cross over into any other state.
