Well the Germans took over Europe without any hassel apart from when they got to Dunkirk. Then came the Battle of Britain and against the Americans was D-Day. The Russians stopped them at Stalingrad.
DUNKIRK:
Wasn't an official battle, just some English and french troops escaping from the beach. Many would say that the Germans lost as they failed their primary target to stop the escapees.
BATTLE OF BRITAIN:
The first major battle the Germans lost against Britain was this one. The Germans flew planes across the channel to bomb RAF bases and cities. They were stopped by RAF planes and were forced to withdraw from the air.
D-DAY
The first loss against the Americans was at D-Day when American, British and Canadian forces landed at Normandy, France. The Germans failed to stop the advances of the troops and pulled back.
STALINGRAD
The Germans first los to the Russians at Stalingrad. The soviets were determined not to lose this city as it had vital oil fields that would make them lose the war if they were captured. This is where heroes like Vasily Zaitsev appeared and snipers became strategic options. The Germans were forced to retreat after they were surrounded and defeated.

Operation Barbarossa was Germany's first major defeat, Germany was hoping by end of 1941 or 194 latest that the USSR would fall and Germany would force britain to submit and possible including america.
