The Articles of Confederation was the first document covering the thirteen colonies.

It did talk about what "constitutes" the American colonies, but it's not technically a constitution. The US Constitution was ratified in 1787.
If you mean "a constitution" not "the Constitution", then the answer is the Articles of Confederation, deemed void by the ratification of the Constitution. The Articles were ill-fated from the very beginning, by the way.
