The normal practice is for the black (or at least darker) wire to be hot. However, given the consequences of guessing wrong, they should be tested and both wires should be treated as "hot" until you find out differently.
If you are asking about the electrical cord on a lamp, the black wire with the white stripe is the neutral conductor.

If your car has a charge indicator light, you should have a white wire with a black stripe hooked to the stator post, a white wire to the fld post, a black wire with a yellow stripe hooked to the bat post, and a black wire with a red stripe hooked to the ground post. The white wire on the fld post goes to the F terminal on the voltage regulator, the white wire with black stripe on the stator terminal goes over to the S post on the voltage regulator, the black wire with the yellow stripe on the bat post goes to the hot side of the starter solenoid, and the black wire on the grd post goes over to one of the attaching screws on the voltage regulator and also to the passenger side of the engine block, where it is attached with the same bolt that holds the negative battery cable to the block. If your car has the charge indicator gauge instead of the light, the stator post is not used.
Any marking (a white stripe, bump molded into the cable, different wire colours) is an indication for your reference only. It doesn't matter, as long as you connect the marked side on the amplifier to the same polarity on the speaker side.

There should be three wires attached to back of the alternator of a 66. You should have a black wire with a red stripe hooked to the GRD post, a black wire with a yellow stripe hooked to the BAT post and a white wire hooked to the FLD post. The black wire with the red stripe is the ground and splits off with one end bolting to the engine block on the same bolt that holds the negative battery cable to the block, the black wire with the yellow stripe goes to the hot side of the starter solenoid, and the white wire goes over to the F post on the voltage regulator.

The orange and green is the 12v feed from the auto shut down relay. The black and white wire is the wire the computer grounds.
White is neutral in home wiring. Red is sometimes used in 3-way switches and dimmer applications, so it is likely red is hot in your application if it pertains to home wiring. The term positive would just apply to DC wiring since AC goes positive and negative. For example your car battery has Red as Positive and Black as Negative.
