It is hard to say for sure, but several of the smaller minicomputers of the 1960s (e.g. DEC PDP-8) could and usually did sit on a desktop. However the term "desktop computer" was not created until the microcomputers of the 1980s and did not come into common usage until laptop microcomputers computers became widely available in the 1990s.

The first was probably the Scelbi in March 1974, followed by the Mark-8 in July, however, the first practical one was the MITS Altair, roughly in January 1975.

This was a bus computer that allowed for many different boards to be plugged into the motherboard. This was available as a kit or a pre-built computer.

The original price for the kit was around $400. The CPU was the Intel 8080, which was an improvement of the 4004, which was created for use in automated gas pumps.

EDIT:completely wrong, Microsoft has no noteable hardware mset-ups, IBM created the first "modern" computers but the first recognized computing machine was designed by Charles Babbage, although the machine was never built in its' time it has been recreated in a musuem in Britain.
