Mecca is the place of the first Quran revelation by Allah (or God and same God in Judaism and Christianity) to Prophet Muhammad through the Angel Gabriel (or Jibril) that started in year 610 AD. Prophet Muhammad (peace be upon him) was born in Mecca (or Makkah) in year 570 AD and He married there with Khadija; his first wife and only wife until she died.
  2. Medina (or AlMadinah) is the second holy Islam city for Muslims because it was the first Islam state and was the focal town from which real spread of Islam started. In addition, there is there the prophet Mosque (AlMasjid Alnabawy). Prophet Muhammad (peace be upon him) died while in Medina (in year 632 AD) and buried there and also the first two prophet successors AbouBakr and Omar.

Makkah, Madina and Jerusalem are the three Holy places for the Muslims. Makkah is the House of Allah - The Kabbah, Madina is the Holy city where Prophet Mohammed (SAW) migrated and lived and died and is buried there, and Jerusalem was the place where the Prophet Mohammed (SAW) prayed with all the other Prophets and ascended to Heaven from the Al Aqsa Masajid. Also Muslims originally used to pray facing the Al Aqsa Masajid, but later the direction was changed and now they face The Kabbah for their prayers.
