Because negative staining requires the use of an acidic stain, which will not penetrate the cells because of the negative charge on the surface of the bacteria. As a result, the unstained cells can be easily identified against the colored background.

Methylene Blue is a positively charged stain it will adhere better to negatively charged cells. Nigrosin is negatively charged so it will be repelled by cells that have a negative charge forming a negative stain. So no methylene is not used as a negative stain as far as i know.

Nigrosin is a stain that can be used to stain microorganisms that are not usually stained by the traditional methods. It is also capable of staining organisms that were not heat-fixed prior to staining. Methylene blue cannot be used in the place of nigrosin because it is not capable of allowing those things to occur.
