Why cant methylene blue be used in place of nigrosin in negative staining?

Because negative staining requires the use of an acidic stain, which will not penetrate the cells because of the negative charge on the surface of the bacteria. As a result, the unstained cells can be easily identified against the colored background.

∙ 13y agoMethylene Blue is a positively charged stain it will adhere better to negatively charged cells. Nigrosin is negatively charged so it will be repelled by cells that have a negative charge forming a negative stain. So no methylene is not used as a negative stain as far as i know.

∙ 13y agoNigrosin is a stain that can be used to stain microorganisms that are not usually stained by the traditional methods. It is also capable of staining organisms that were not heat-fixed prior to staining. Methylene blue cannot be used in the place of nigrosin because it is not capable of allowing those things to occur.
