The Speaker of the US House of Representatives (currently John Boehner) is the immediate successor to the Veep.
After the Speaker in order follow the President Pro Tempore of the Senate, Secretary of State, Secretary of Treasury, Secretary of Defense, Attorney General, Secretary of the Interior, Secretary of Agriculture, Secretary of Commerce, Secretary of Labor, Secretary of Health and Human Services, Secretary of Housing and Urban Development, Secretary of Transportation, Secretary of Energy, Secretary of Education, Secretary of Veterens Affairs, Secretary of Homeland Security. (The cabinet officers are ranked by the order in which their departments were created.) If the person holding any of the positions is not legally eligible to be President, he is skipped over.
Please understand that this order would only apply in some almost unthinkable emergency that created a large number of vacancies at the same time. In most cases, a vacancy in the vice-presidency would be quickly filled by presidential appointment and confirmation by Congress.
