I don't know if/how it can be done in Assassin's Creed 2, but in the first Assassin's Creed you can kill anyone without penalty after completing the game and replaying story mode. This site might make that a bit more understandable:

It is Desmond you are talking about. The menu is different, and you can't do different memories. When in the Animus, you can. Otherwise, you can't go back to memories when Desmond.

I'm sure if you had the subtitles switched on during the game, you could do a translation search on their meaning. All I know of the top of my head that makes sense is when people say "pazzo" it means they think I'm crazy. Hearing people saying a rhyming word beginning with C is quite possibly the most widely used crude word in the world. It also means "penis". Finally, hearing Ezio repeatedly quote the words after a kill "requestio de pace" means "rest in peace". This was all by logical surmisement on my part, but it would be interesting to learn more Italian and play it in the native tongue for authenticity. Arrivederci is also heard several times in the game. It means good-bye

I don't think there's a specific reason you can't get there, the developers at Ubisoft just didn't see any need to extend the city further and add in that much more work, so they left it.
