Not unless there is a cut-away in the body. For most ukuleles it is just a matter of moving the strings around. A few of them may have a 'pick guard' even though most players don't use a pick. Some electric ones have a pick guard, mostly to make it look like a little guitar, as said, a pick is not typically used on a uke.
Most people, regardless of their orientation, strum with the right and do the chords with the left. Most teachers recommend doing that as it means tabs can be used as is without having to flip everything around.
And in mass production, they don't make a left handed version. Most luthiers would be able to create a custom made one with the reverse cut outs if someone really wanted it.

It could very well be the tuning pegs. It could also be the strings.
At the end of your tuning pegs there should be screw heads. Use a properly sized screwdriver to tighten those up just a bit. Be careful! You do not want to over tighten! But that increases the friction and should help hold the tuning better.
If the strings are fairly new, they have to 'stretch' into a point of equilibrium. If this is the case, it should settle out after a couple weeks. Temperature swings can also have an effect.
