Both bacteria types would be stained by the safranin. When the iodine is added, safranin would be "set" in the positive. The decolorizer would wash out the safranin and then application of the crystal violet would stain the negative.
During gram staining, if in the application process crystal violet was reversed with safranin by accident. The safranin would be washed out and/or covered up by the darker crystal violet dye.
Then, there might be an inaccurate result or might get wrong in reading the possible bacteria present in the specimen.
Gram positive bacteria will remain violet, but without the counterstain gram negative bacteria will not be the recognizable pink/red color they need to be in order to classify the organsim.
gram-negative bacteria will be clear or colorless
Endospores are impermeable to most stains so heat is usually applied to drive the stain into the endospore.
If you stop at this stage (without the alcohol), the Gram-negative cells will be invisible since they have lost their crystal violet stain.
It acts as a fixative and fixes the cells
The carbolfuchsin is a differential stain. It is an acid-fast stain which distinguishes organisms with waxy cell walls with the use of acid alcohol.
It is an alchoholic compound i.e. 95% ethanol.
If used sparingly, especially in cases of using rubbing alcohol to clean a small stain, rubbing alcohol evaporates quickly enough to not cause damage to microsuede furniture.
Rubbing alcohol all over the stain
no.
To make stain resistant paper a special Teflon coating is applied to the paper. This coating can be applied to all types of paper products.
If alcohol (decolorizing step) is omitted then the primary stain absorb by the bacteria will not be washed away. This will result in all or nearly all the bacteria to appear purple in color under the microscope.
The decolorizing agent in the acid fast stain is acid alcohol. The decolorizing agent in the gram stain is ethanol.
When the first is cured. Usually 2-3 days.
Try using vodka to clean it. Vodka is clear and should be able to dissolve the stain.
yes isopropyl alcohol stain the plastic? interior of my car (Toyota Fortuner and i can not remove this White like decloration if any body Know how to remove this stain please let me Know i will apreciate it Armando Pérez armandoapp@gmail.com
