For the 3.5L Engine it is on the top rear part of the engine facing towered the cabin. You will see the thick black hose coming out of it.
On the 2.8 it's on top of the plenum at the back. disconnect the hose & unscrew the pcv valve & replace.
On 2.7L engines the PCV valve is threaded into the intake plenum above the throttle body. On 3.2/3.5 engines it is on the rear drivers side of the plenum. Both engines have hoses attached going to the drivers side valve cover.

it's in the intake plenium, in the rear center. Its black plastic and has a hose attached to it that goes into the valve cover.
Attached to the valve cover is a hose on the driver side. Unscrew the nut closest to the valve cover and that is the PCV valve. It is a straight PCV valve, not the elbow shape you may be looking for.
It is threaded into the valve cover. There is a rubber tube that attaches to the valve cover - pull off the tube (there may be a clamp) - then unscrew the PCV valve with a wrench.

Some of the 2000 models have a PCV valve on the back of the valve cover. The other models do not have a PCV valve, but use an oil/air separator instead.
The PCV valve is located in the valve cover, on the 4.7L it is in the side of the oil filler housing.
The PCV valve for a 2000 Lexus rx300 is on the passenger side of the engine. It is on the valve cover near the back.
