Queen Isabella of Spain appeared on the commemorative quarter distributed at the Chicago Worlds Fair in 1893. A half dollar featuring Christopher Columbus was also issued. These coins were sold for $1 apiece but it seems everyone in those days must have been bargain hunters. Many people were willing to pay double face value for the half dollars but few would pay 4 times the value of a quarter, so today the quarter is much scarcer and more expensive than the half.
Excluding the various images of "Liberty", the first woman on US coinage was Susan B. Anthony, on the 1979 One Dollar Coin. That being said, some of the images of "Liberty" were designed using real models. For example, The Peace Dollar was designed by Anthony de Francisci, who may have used his wife as the design model. However, since this is portrayed as one of the many images of "Lady Liberty", the identity of the models used does not constitute an honor bestowed upon an individual. Experts and Historians agree that this honor (first woman on US coinage) does, in fact, belong to Susan B. Anthony.
Other non-allegorical images since then have included:
> Sacajawea, on $1 coins struck since 2000. However there is no known picture of her actual appearance, so the designer (Glenna Goodacre) used a Native American student as her model.
> Helen Keller, on the reverse of the Alabama state quarter.
