Yes Mary of Scotland was a threat to Elizabeth because Mary was a catholic the catholic people wanted Mary to be queen of England so England can become a catholic country . Also Mary of Scotland was a successful queen and she provided heir to he throne whilst Elizabeth refused to marry and provide heirs to the throne

She wasnt the next in line to the throne after Elizabeth, but she was close. The reason for this was because she had Tudor blood in her. Her grandmother Margret was the sister of Henry VIII,and the daugher of Henry VII. Also she was married to someone who had strong claims to the throne of England as well. Her husband was her first cousin Henry Steward, Lord Darnley (they had the same grandmother but different grandfathers.) That is why their son James became King after Elizabeth died.
