You have to refill through the dip-stick tube, requires a special funnel. Check your owners manual for capacity probably about 10 quarts. Check the owners manual for the location of the transmission dip-stick and be sure you don't put any into the oil dip-stick port. The dip-sticks are usually different colors, the transmission are typically red. BAD ANSWER == There is no dip stick on a 2006 Ford Explorer . . . it is a sealed transmission. Remove said plug. Screw in a brass fitting with 1/8 X 28 NPT on one end and 3/8 hose nipple on the other. attach 3/8 rubber hose from nipple to a funnel( i used a suction gun from a local parts store, take it apart and you can use the plunger to force feed). support the funnel above the level of the transmission and let it gravity feed.
There is no drain plug assuming it is an automatic transmission it is drained when removing the transmission pan and changing the transmission filter.

The automatic transmission fluid should be changed every 30,000 miles. The transmission oil is checked under the car, pull plug to drain into pan, once drained reinsert plug and fill via fluid pump until the level bottom of the hole.

Automatic transmission: Hyundai SP-III ATF. 8.2 quarts or 7.7 liters based on filling the system from totally empty. Replacement volume would best be based on the volume drained during change-out.
I suggest you have the fluid completely drained and a new filter installed. Do not have the system flushed. You will only stir up trouble. Use Mercon or Mercon V automatic transmission fluid. Ford says that all applications that previously required Mercon may now be serviced with Mercon V.
If it is completely drained and flushed, about 9-10 quarts. If you just drained the transmission pan from the drain plug, about 4 quarts.
If you drained the pan, it will take about 4-4 1/2 qts to fill it. If you drained the whole transmission it will take more like 8 or 9.
